Description | Polymyxin E, known as colistin, is a narrow-spectrum bactericidal antibiotic of the lipopeptide group, effective against Gram-negative pathogens. In clinical practice, it is administered as a prodrug colistin methansulfonate (CMS), which is spontaneously hydrolysed to colistin. CMS stability is dependent on many factors such as concentration, pH, temperature, and composition of the carrier solution. The quantification of CMS is based on indirect determination, which is burdened with several problems. Correct and accurate measurement of colistin and CMS concentration is important to find the optimal dosing regimen, to reduce drug toxicity, and contributes to the understanding of stability and pharmacokinetics of the drug and prodrug. The aim of this work is to highlight the key points in the determination of colistin and its prodrug by LC-MS. |
