Description | Molecular biological methods based on genus and species specific PCR and restriction analysis of two PCR amplicons using six restriction enzymes were used to differentiate among species of Bifidobacterium. Optimalised methods were applied for identification of strains collected in Culture Collection of Dairy Microorganisms. Restriction analysis of longer PCR amplicon was the most discriminative for Bifidobacterium species differentiation. Practicaly observed restriction profiles were compared and confirmed with the theoretical restriction analysis of Bifidobacterium sequences obtained from GenBank database. |
