Description Abnormal expression of cell cycle regulators, i.e. D1 cyclin, and cyclin dependent kinase inhibitor, i.e. p16 protein, are routinely detected in many types of primary tumors. However, the significance of the above regulators as predictive factors in carcinomas of the orofacial region has not been studied yet. For this purpose a retrospective database (2000-2005) of 68 patients was compiled, in whom the resection of spinocellular carcinoma of either the tongue or the oral floor, or, possibly, the block dissection of the neck were carried out. Immunohistochemical examinations for p16 and D1 protein demonstations were carried out on selected blocks of routinely fixed (4% formol) and treated material. The results were evaluated in a semi-quantitative way; the degree of expression (increased D1 expression and/or p16 deletion) was compared with a number of clinical-pathological data (age, sex, grading, staging, diseasefree period, survival time, and meatastases). The aim of the study was to confirm the correlation between reduced p16 expression and/or cyclin D1 over-expression and a worse prognosis of the tumorous disease including the development of node metastases. The outcome of the study corresponded to data published recently. Evaluation of expression of the above cell cycle regulators enables to define the subgroup of patients with higher risk for loco-regional recurrence and tumor diffusion into regional lymph nodes, which has imminent affect on disease conclusion and patient's survival.
