Description It may have been shown that the deviating structure of the glucan net fromed by protoplasts of Sacch. cerevisiae in a liquid medium, as compared with the glucan net in normal walls of this species, is explainable solely from a deficiency of beta-1,6-synthetase effected by the mediun and the resulting change in constitution and physical properties of the microfibrils. This explanation implies that the fibrils are regarded as products of directed organization and their fascinations as products of self-assembly. The type of net discussed appears to be formed in a liquid medium also by protoplasts of other species of yeast, namely Schizosaccharomyces pombe and Nadsonia elongata. But the fascination of microfibrils do not reach the width sometimes observed with Sacch. cerevisiae and later these species form a complete wall bellow the initially formed net. The cell wall of the genera in question is essentially different in constitution from the Sacch. cerevisiae, and apparently the matrix material does not as easily diffuse in the medium as with this species. However, in both Schizosaccharomyces and Nadsonia the wall contains an alkali-insoluble glucan which, like that of Sacch. cerevisiae, yields microcrystalline beta-1,3-glucan on boiling with dilute acid. The wide meshed nets of long microfibrils formed during the first period of culture suggest therefore that regeneration starts with glucan synthesis in which a deficiency of branching factor caused by the medium induces synthesis of long, unbranched chains.
