Description | The work deals with the electrochemical study of three short homo–deoxyoligonucleotides (homo–ODNs) containing cytosine (C). Using linear sweep voltammetry (LSV) and elimination voltammetry with linear scan (EVLS) the effect of pH and the number of C in the ODN chain on reduction signals of dC3, dC6, and dC9 in buffered and non-buffered solutions was investigated. Reduction C peaks, recorded at mercury electrode in the potential range from –1.1 V to –1.7 V vs. Ag/AgCl/3M KCl, are strongly dependent not only on pH but also on the composition of solutions. With the addition of buffer (acetate-phosphate buffer) to 0.1 M NaCl the reduction peaks increase and move to more positive potentials. In non-buffered solutions changes of voltammetric responses of all ODNs in time were observed. The biggest changes were recorded in the case of dC6. The adsorption or surface kinetics of ODNs was confirmed by EVLS and electrochemical behaviour of ODNs studied was discussed in the context of the i-motif formation. |
