Description | Guanosine derivatives are important for diagnostic of oxidative DNA damamge. 8 - hydroxy - 2-deoxyguanosine (8-OHdG) is one of most abundant products of DNA oxidation. Its determination in biological fluids, e. g. urine is possible. Nevertheless, owing to its low concentration and matrix interferences (uric acid) use of HPLC without previous extraction is difficult. In this study we tested the isolation of 8-OHdG from a biological matrix by using the paramagnetic particles with antibody - modified surface. We used the sandwich ELISA principles for determination of molecules of 8-OHdG. The primary antibody was bound to a paramgnetic particle, then 8-OHdG was attached during the reaction in the sample. Then the secondary antibody with alkaline phosphatase was attached. Concentration of the analyte was determined from concentration of napthol, which is a product of alkaline phosphatase where napthyl phosphate acts product.
