Description | Rationale: Human induced pluripotent and embryonic stem cells (hiPS and hES) can be differentiated into cardiomyocytes (CMs) in vitro. Developed methods like cocultivation with END2 cells, differentiation on matrigel and also cytokine effect on clusters of cells, so called EB method. The current studies are facing very low yield and process variation according to the type of used stem cell line. Objective: The aim of this work is to generate human CMs from AggreWell plates through EB method. Methods and results: Cardiomyocytes were generated through modified protocol inspired by Gordon Keller s lab work(Toronto, Can). Subsequent analyzes of gene expression, imunohistochemistry and electrophysiology proved cardiomyocyte – like character of differentiated cells, neverthless the cells represent very young stage of developing heart and has to be further investigated. This basic research opens way for further pacient specific investigation on the level of cellular cardiology. |
