Popis | We identified the cytoskeleton in Cryptococcus neoformans, Aureobasidium pullulans, Fellomyces fuzhouensis, Cryptococcus laurentii and Malassezia pachydermatis. The cytoskeleton is involved in budding, bud-fission, hyphal growth and conidiogenesis of human yeast and fungal pathogens. We investigated the cytoskeleton as a potential target for inhibition of morphogenesis using microtubule and actin inhibitors. All microtubule inhibitors were effective, but clinical ones only transiently and resistant cells originated. Cytochalasins were not effective. Latrunculin A inhibited budding, fission, hyphal growth, conidiogenesis. The most efficient inhibition was reached by combining the microtubule and actin inhibitors (Kopecká M.,Gabriel M., Chemotherapy 2009;55:278-286). Our study provided new results of the basic research on the cytoskeleton as a new potent target for inhibition of yeast pathogens that may show the way to a new antifungal strategy. |
