This project proposal investigates various aspects of biooxidation of inorganic sulfur substances using acidophilic bacteria in relation to biochemical and chemical processes taking place in sulfide wastes and other deposits. Environmental acidification and metal migration are the results of those processes. Biooxidation of sulfur substances will be studied at cell and enzyme levels. Mechanism of pyrite biooxidation will be investigated in relation to a possible formation of sulfur intermediates during the indirect oxidation process. Inhibition of elemental sulfur biooxidation by arsenic will be studied due to the arsenic occurrence in sulfide minerals (arsenopyrite) to learn its inhibitory mechanism and environmental consequences. Acidithiobacillus ferrooxidans bacteria were isolated in the past as a dominant organism in sulfide waste at Zlaté Hory participating in water acidification. The heap surface has been recultivated. The inner heap layers will be a subject of testing to learn the effect of recultivation on the occurrence of active bacteria which could take part in further environmental acidification.
